I have no sound daemons running, so this was an easy test with ALSA
0.9.0Beta7.

I did the steps described and play executes perfectly.  In fact, I took
it one step further, started the .mp3 with xmms and then executed the
play command and had both sounds playing at once!  Tested with both a
.wav and .au file.

So it would appear that with Linux 2.2.19 and ALSA, your notification
would play right through the mp3.
